Mutation of the glutamic acid residue at position 355 to glutamine reduced
the Kcat of the enzyme but not the Km2. Kcat is
equal to Vmax/concentration of enzyme3. The
terminology E355Q means that Glutamate (E) at position 355 has been mutated
to glutamine (Q)4. The
mutation E355Q caused the pH optimum to shift to more basic pH5. With
reference to table 3 the fact the mutations affected the Kcat much more than
the Km indicate that while the altered amino acid are important for the
catalysis of reaction they are not as important for maintaining the stability
of the tertiary structure of the enzyme
